Pseudochrobactrum albidum sp. nov., a dimethylformamide-degrading bacterium isolated from activated sludge of a wastewater treatment plant

Abstract

A Gram-stain negative, aerobic, oxidase-positive, non-motile, rod-shaped bacterium (approximately 1.0–1.5 µm in length and 0.4–0.6 µm in width), designated HB0163ᵀ, was isolated from activated sludge of a wastewater treatment plant in Hangzhou, Zhejiang Province, China. The strain demonstrates the capability to efficiently degrade N, N-Dimethylformamide in industrial wastewater. Strain HB0163 T grew at 20–45°C (optimum 35°C), in 0–6.0% (w/v) NaCl (optimum 2.0–3.0%) and pH 5.0-9.5 (optimum 7.5-8.0). Chemotaxonomic analysis indicated that the respiratory quinone was Ubiquinone-10, the dominant cellular fatty acids were C 19:0 cyclo ω8c, C 18:1 ω7c and C 16:0 , and the major polar lipids were phosphatidylcholine, phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylmonomethylethanolamine, two unknown phosphoglycolipids, three unknown aminolipids, two unknown phospholipids, and one unknown lipid. The DNA G + C content of strain HB0163 T was 50.7% based on its genome sequence. Phylogenetic analysis based on 16S rRNA gene sequence indicated that strain HB0163 T formed a distinct lineage in the clade of the genus Pseudochrobactrum . Strain HB0163 T also exhibits low digital DNA-DNA hybridization (< 70%) and average nucleotide identity values (< 98%) with other Pseudochrobactrum species. According to the polyphasic taxonomic characterizations above, strain HB0163 T is considered to represent a novel species of genus Pseudochrobactrum , for which the name Pseudochrobactrum albidum sp. nov. is proposed. The type strain is HB0163 T (= MCCC 1K09177 T  = JCM 37008 T ).
