Omega-3 fatty acids plus aloe-emodin prevents alveolar bone loss in a rat model of periodontitis

Objectives : The aim of this study was to evaluate the effects of omega-3+aloe-emodin (AE) on alveolar bone in rats with periodontitis induced by porphyromonas gingivalis. Materials and Methods : The experimental rats were randomly divided into six groups (n=5 each): healthy control, disease control, vehicle control, omega-3 plus AE treatment group, AE treatment group and omega-3 treatment group. After all maxillary second molars in G2-G6 ligated 7 days, followed by daily treatment of G4-G6 with omega-3 only, AE only and omega-3+AE for 14 days. All rats were sacrificed at day 21. RAW 264.7 cells were cultured with lipopolysaccharide and receptor activator (NF)-κB ligand and added with omega-3, AE and omega-3+AE. Then, the inflammatory factors and osteoclastogenic gene were measured. Results : Omega-3+AE successfully inhibited alveolar bone loss and promoted alveolar bone growth. In vitro experiments showed that omega-3+AE also inhibited inflammatory factors and osteoclastogenic gene. In addition, omega-3+AE enhanced the expression of the osteoblast specific marker protein. Conclusions : These results suggests that omega-3+AE could reduce porphyromonas gingivalis-induced alveolar bone loss in rats with periodontitis. Clinical Relevance : These results indicate that omega-3+AE can inhibit the destruction of periodontal tissue caused by periodontitis and improve the quality of life, which has important clinical significance and wide application prospect.
