Abstract

The synthesis of Ag NPs using Rhazya stricta leaf extract, rich in phytochemicals, is explored for its potential in cancer therapy. Ag NPs were successfully synthesized at pH 8 and temperatures of 60°C, 70°C, and 80°C, exhibiting a characteristic SPR peak at 407 nm. SEM analysis revealed spherical nanoparticles (68±1 nm), suitable for biomedical applications. The synthesized Ag NPs demonstrated potent anticancer effects against MCF-7 breast cancer cells, surpassing the leaf extract alone, with low LC50 values. Mechanistic insights suggest apoptosis induction, hindered cell proliferation, ROS generation, mitochondrial disruption, and interference with cell signaling pathways. This study underscores the potential of Rhazya stricta-mediated Ag NPs as promising anticancer agents, contributing to the optimization of synthesis conditions and expanding our understanding of their multifaceted mechanisms in cancer therapy.
