A green digestion method for biological samples with hydrogen peroxide as the main digestion solvent

The present study examines an environmentally friendly, efficient and safe digestion method for biological samples using hydrogen peroxide (H 2 O 2 ) as the primary digestant, aiming to replace the widely used traditional digestion methods based on concentrated acids such as nitric acid (HNO 3 ), hydrochloric acid, and sulphuric acid (H 2 SO 4 ). Initially, the concentrations of H 2 O 2 and HNO 3 were optimised, followed by adjusting factors such as pressure, temperature and time to establish a heating programme that ensures uniform and complete digestion of various representative animal and plant tissues under mild conditions. Under low-pressure conditions, a digestion solution containing 8%–16% H 2 O 2 and 3%–4% HNO 3 effectively digested various biological materials, yielding excellent outcomes. Based on a comprehensive ecological scale analysis, this method exhibits advantages in terms of environmental friendliness, safety, cost-effectiveness and high efficiency, ultimately achieving a total score of 85, fully validating its potential and value as an outstanding green digestion method.
