Synergistic Effects Of Platelet-Rich Fibrin And Photobiomodulation On Migration Of Mg-63 Pre osteoblatic cell line: An In Vitro Study

Objectives: To evaluate the synergistic effect of L-PRF and Low-level laser therapy on the migration of Mg-63 pre-osteoblastic cells. Settings and Design: In- vitro comparative study Methods and Material: L-PRF was prepared from peripheral blood obtained from six healthy individuals using Choukroun’s protocol. Conditioned media (20%) obtained from L-PRF membranes was utilized in a Transwell migration assay. MG-63 cells were divided into three groups: control (2% FBS), PRF-treated, and PRF combined with LLLT. The PRF+LLLT group was exposed to diode laser irradiation (810 ± 10 nm, 6 J/cm², 60 seconds) in a non-contact, non-pulsed mode. After a 24-hour incubation, cells that migrated to the lower membrane surface were fixed, stained with 0.5% crystal violet, and quantified using ImageJ software. Statistical analysis : Statistical analysis was performed using one-way ANOVA with Tukey’s post hoc test. Results: PRF treatment alone resulted in a 92% increase in osteoblast migration when compared to controls (p < 0.001*). The combined application of PRF and LLLT produced a 184% increase in migration compared to the control group (p ≤ 0.004), indicating a statistically significant synergistic effect. Conclusions: The findings demonstrate that synergistic use of PRF and LLLT substantially increases osteoblast migration, suggesting their potential for combined use in clinical bone regeneration strategies.
