Blunt End Ligation in Molecular Cloning

Molecular cloning is a set of experimental methods in molecular biology used to assemble recombinant DNA molecules and direct their replication within host organisms.,the most used method was sticky end ligation,here we creat a new blunt end ligation without PCR again, Blunt ends are created when restriction enzymes cut DNA without leaving overhangs, resulting in straight, double-stranded ends.we found a way to improve ligase activity and successful ligation,the most thing in clonning is make sure the ligation product can work and produce protein successfuly,we also show the protein photo to demonstrate its work.This article is demonstration blunt end clonning,you can select any gene in same way,we used Histone synthetic lethal7 gene as a target gene,you just need to find vector and target restrict site then you can conduct experiments.i hope this article can help people who need to do molecular cloning.
