Abstract

In humans, ACE2 and TMPRSS2 are expressed in various cells and tissues, and differential expression has been described in young and old people, with a higher level of expressing cells being detected in the nasal brushing of older people than young individuals. We described the same pattern occurring in ferrets, and we demonstrated that age affects susceptibility of ferrets to SARS-CoV-2.

EthicsIACUC: Animals housing and experimental design: All animals were handled in accordance with the Animal Welfare Act and the study procedures were reviewed and approved by the Institutional Animal Care and Use Committee at the Cornell University (IACUC approval number 2020-0064).
Euthanasia Agents: Ferrets were humanely euthanized on day 14 pi under deep inhalatory anesthesia using isoflurane followed by heart puncture.
Sex as a biological variablenot detected.
RandomizationTwenty young (6-month-old ferrets) and twenty aged ferrets (18 to 39-month-old [30.5±5.3 - average±SD]) were randomly allocated to experimental groups (n = 4).
BlindingCell cultures with no CPE were frozen, thawed, and subjected to two additional blind passages/inoculations in Vero E6/TMPRSS2 cell cultures.
    Power Analysisnot detected.
    Cell Line Authenticationnot detected.

    About SciScore

SciScore is an automated tool that is designed to assist expert reviewers by finding and presenting formulaic information scattered throughout a paper in a standard, easy to digest format. SciScore checks for the presence and correctness of RRIDs (research resource identifiers), and for rigor criteria such as sex and investigator blinding. For details on the theoretical underpinning of rigor criteria and the tools shown here, including references cited, please follow this link.
