Abstract

The Manouria impressa (Impressed tortoise) is a unique fungivore species with restricted distribution in Southeast Asia, recognized for its evolutionary importance and conservation priority. Here, we present the first de novo genome assembly of an impressed tortoise sampled from Yunnan, China. Using PacBio HiFi long-read sequencing and Hi-C proximity ligation technologies, we generated a 2.29 Gb assembly comprising 81 scaffolds with a contig N50 of 78.37 Mb, a scaffold N50 of 148.31 Mb, and a BUSCO completeness score of 98.3%. This high-quality genome assembly provides a critical resource for exploring the genetic basis of evolutionary adaptations and informing conservation genomics to support the management of this vulnerable species.
